Overview

Neurotensin is a 13-amino acid neuropeptide widely distributed in the central nervous system and gastrointestinal tract, where it modulates dopaminergic neurotransmission, nociception, and intestinal function. It acts primarily through the neurotensin receptor 1 (NTSR1), influencing mesolimbic and nigrostriatal pathways implicated in motivation, reward, and motor control.

Neurotensin has been studied for its potential roles in schizophrenia, pain modulation, and cognitive function.

Research Summary & Key Findings

Preclinical studies have demonstrated that neurotensin receptor agonists can modulate dopamine release and influence behavioral models of psychosis and cognition, with publications in neuropsychopharmacology journals over the past three decades. Altered neurotensin levels have been observed in postmortem brain tissue from individuals with schizophrenia. No neurotensin-based therapeutics have reached clinical trials due to poor stability and CNS penetration of the native peptide.
