Tabimorelin
Overview
Tabimorelin is an orally active synthetic ghrelin mimetic and selective GHS-R1a agonist developed to stimulate growth hormone release. Unlike peptide analogues, tabimorelin was designed as a small molecule with improved oral bioavailability and stability.
Mechanism of Action
The therapeutic rationale centered on treating frailty, functional decline in the elderly, and conditions associated with growth hormone deficiency.
Research Summary & Key Findings
Phase 2 clinical trials evaluated tabimorelin in elderly hip fracture patients and in sarcopenia, with results published in the Journal of Clinical Endocrinology and Metabolism in 2007 showing increases in growth hormone and IGF-1 levels. Development was discontinued before Phase 3 completion, and no regulatory approval was pursued. Evidence supported hormonal activity but did not establish clinical benefit in functional outcomes.
Clinical Status
Tabimorelin is in the investigational stage. While preclinical and early-phase data exist, it has not received regulatory approval for clinical use in the United States.
