Matrixyl Synthe'6 (Palmitoyl Tripeptide-38)
Overview
Palmitoyl tripeptide-38 is a synthetic lipopeptide designed to stimulate synthesis of six major constituents of the dermal matrix: collagen I, III, and IV, fibronectin, hyaluronic acid, and laminin 5. It is proposed to activate transforming growth factor beta (TGF-beta) signaling in fibroblasts, thereby promoting coordinated extracellular matrix assembly.
Mechanism of Action
The palmitic acid conjugate enhances dermal bioavailability when applied topically.
Research Summary & Key Findings
In vitro studies using cultured human dermal fibroblasts have demonstrated dose-dependent increases in collagen I, III, and IV mRNA expression, along with elevated hyaluronic acid production. A small manufacturer-supported clinical trial reported volumetric improvements in nasolabial fold depth after 60 days of topical use, though peer-reviewed validation is lacking. Histological confirmation of matrix remodeling in human skin has not been extensively published.
Clinical Status
Matrixyl Synthe'6 (Palmitoyl Tripeptide-38) is in the investigational stage. While preclinical and early-phase data exist, it has not received regulatory approval for clinical use in the United States.
