FOXO4-DRI
Overview
A D-retro-inverso peptide designed to disrupt the interaction between FOXO4 and p53 in senescent cells. In senescent cells, FOXO4 sequesters p53 away from mitochondria, preventing p53-mediated apoptosis and enabling senescent cell survival.
Mechanism of Action
FOXO4-DRI competitively binds p53, releasing it to trigger selective apoptosis of senescent cells while sparing healthy cells, functioning as a senolytic agent.
Research Summary & Key Findings
In vivo studies in naturally aged and fast-aging (XpdTTD/TTD) mice demonstrated restoration of fitness, fur density, and renal function after FOXO4-DRI treatment (Baar et al., Cell, 2017). Selectively induced apoptosis in senescent cells in vitro without affecting proliferating or quiescent cells. Preclinical stage with no human clinical trials initiated. The D-retro-inverso design confers protease resistance for improved in vivo stability.
Clinical Status
FOXO4-DRI is in the research phase with limited clinical data in humans. Current evidence is primarily derived from preclinical (animal or in vitro) studies.
