FOXO4-DRI

01 · Overview
A D-retro-inverso peptide designed to disrupt the interaction between FOXO4 and p53 in senescent cells. FOXO4 helps senescent cells avoid apoptosis by binding p53; the FOXO4-DRI peptide perturbs this interaction, selectively causing p53 nuclear exclusion and cell-intrinsic apoptosis in senescent cells (PMID 28340339).
Structural studies show FOXO4-DRI binds the disordered transactivation domain of p53 (PMID 40593617). The D-retro-inverso design is intended to resist protease degradation.
02 · Research summary & key findings
In fast-aging XpdTTD/TTD mice and naturally aged mice, FOXO4-DRI restored fitness, fur density, and renal function, and it neutralized doxorubicin-induced chemotoxicity under well-tolerated conditions (Baar et al., Cell, 2017) (PMID 28340339). Its senolytic effect is selective for senescent cells, inducing apoptosis through the FOXO4-p53 axis (PMID 28340339). Evidence is limited to cell and animal studies; no human clinical trials have been reported.
03 · Regulatory status
FOXO4-DRI is in the research phase with limited clinical data in humans. Current evidence is primarily derived from preclinical (animal or in vitro) studies.
No lawful pathway today
FOXO4-DRI is not FDA-approved and does not currently have a lawful prescription or compounding pathway in the United States, so we do not sell it and will not book a consultation for it. That can change: in 2026 FDA began reviewing several peptides for compounding.
